Yes. An ECM rash is diagnostic for Lyme disease. Current guidelines approved by ILADS (International Lyme and Associated diseases Society) recommends oral therapy for at least 6 weeks for both adults and children when a rash is present. Lyme patients who are pregnant have special guidelines to help protect themselves and their unborn child. According to the CDC, Lyme disease is a 'clinical diagnosis' and negative tests are not to be used to rule out the disease.
